Achttien jaar na de eerste missie van God van de Blues Brothers komt Elwood Blues vrij en ontdekt dat er al die jaren veel is veranderd. Zijn maatje Jack is dood, zijn band is uit elkaar en het weeshuis waar hij is opgegroeid, is gesloopt. Elwood ontdekt al snel dat hij een nieuwe missie te volbrengen heeft: hij moet de band opnieuw bij elkaar halen, ditmaal met de hulp van een barman vol soul, hij moet meedoen aan de Battle of the Bands bij Queen Moussette en hij moet een nogal eigenwijs weeskind genaamd Buster op het juiste pad brengen. Temidden van alle chaos moet hij de politie bewijzen dat er magie in zijn muziek zit... en dat er achter de Blues Brothers-gekte een mysterieus en uniek recept schuilt.
